چکیده

Using immunological techniques we tried to determine the localization and orientation of the sulpholipid of chloroplasts in thylakoids. During immunization of rabbits with lamellar systems of chloroplasts of A n tirrh in u m m ajus antisera were obtained which precipitated sulpholipid emulsions. Antibodies against the sulpholipid were not detected until a second series of intravenous injections. By immunizing purified sulpholipid of F ucus serratus, adsorbed to methylated bovine serum albumin, sulpholipid antisera were acquired. These antisera and the antisera against lam ellar systems of A n tirrh in u m m ajus reacted with preparations of sulpholipid from widely different plant species. The sulpholipid antisera did not react with emulsions of the monoor digalactosyl-diacyl glycerol, of lecithin or the anionic phosphatide phosphatidyl glycerol. Lamellar systems isolated without osmotic shock on a sucrose gradient were not agglutinated by sulpholipid antisera directly, but an indirect agglutination could be achieved in a C o o m b s -test or in a mixed antigen aggluti nation after U h l e n b r u c k . Broken chloroplasts, however, were agglutinated directly by sulpho lipid antisera, as well lam ellar systems which had been disrupted in a French-press or by ultrasonic treatm ent. From agglutination and agglutination inhibition experiments with lamellar systems prepared by different methods it is concluded that the determinant groups of the sulpholipid are accessible for antibodies in the lam ellar system of chloroplasts. But probably the sulpholipid molecules are located inside the thylakoids.

Binding of antibodies onto the thylakoid membrane. I. Maximal antibody binding and adsorption of antibodies to lipids.

The binding of antibodies onto the lamellar system of Antirrhinum majus was determined in dependence on the serum addition. The unspecific adsorption of serum proteins was taken into account or eliminated. The binding of antibodies as a function of the amount of serum added is seem from a saturation curve. From an antiserum obtained by hyperimmunization with stroma-freed chloroplasts, the chlor...

Use of heterophilic agglutinins in plant serology.

In previous communications it has been demonstrated that monogalactosyl diglyceride and the anionic chloroplasts lipids can be detected on the thylakoid membrane by specific antisera 1 _ 3 . The antigen determinants are of carbohydrate nature as was shown by specific agglutination inhibition tests. They are located on the outer surface of the thylakoid membrane and are directly accessible to th...

Localization of the tri- and digalactosyl diglyceride in the thylakoid membrane with serological methods.

Trigalactosyl diglyceride was isolated from leaves of Urtica dioica and characterized by thin layer chromatography, infrared spectroscopy and by its fatty acid composition. An antiserum to the trigalactolipid was obtained by immunization of rabbits. By means of inhibition experiments with oligosaccharides and mono- and digalactosyl glycerol it was demonstrated that the antibodies are directed t...

Binding of antibodies onto the thylakoid membrane. IV. Phosphatides and xanthophylls in the outer surface of the thylakoid membrane.

Antisera to the phosphatides phosphatidyl inositol and phosphatidyl choline as well as to the xanthophylls violaxanthin and zeaxanthin were obtained by immunization of rabbits. The phosphatidyl choline antiserum did not give cross reactions with thylakoid membrane glycolipids, nor with phosphatidyl glycerol and phosphatidyl inositol. The antiserum to phosphatidyl inositol also did not react wit...

Peroxisome dependency of alkyl-containing GPI-anchor biosynthesis in the endoplasmic reticulum.

Glycosylphosphatidylinositol-anchored proteins (GPI-APs) play various roles in cell-cell and cell-environment interactions. GPI is synthesized in the endoplasmic reticulum (ER) from phosphatidylinositol (PI) through step-wise reactions including transfers of monosaccharides and preassembled GPI is transferred en bloc to proteins. Cellular PI contains mostly diacyl glycerol and unsaturated fatty...
